This is very effective product for every aspect of analysis either drug assay or content uniformity or impurities testing. Reproducibility and repeatability results always found within 1% RSD. 21 CFR compliant system with Empower 3 software. System is very robust and rugged and minimum service required for repairing and maintenance.

The instrument has user-friendly features that makes the user to effectively utilize its capability and technicality to expedite method development activity. Water's HPLC system connected with empower software has much advantage in terms of operation, acquisition of data, review and processing of data is much easier and faster. The system has salient features with robust pumping mechanism that delivers accurate gradient composition of mobile phase and hence, accuracy of gradient methods is not compromised. The sensitivity of instrument is good and thus support for the estimation of impurities present at lower level with precision and good repeatability. Reproducibility from one system to another system is excellent. The another advantage is percentage of relative standard deviation for replicate injections of standard solution which is as low as less than 0.7 %.This feature supports users to meet current United states pharmacopeia recommended limits. The additional feature of injector mechanism and design helps lot in reducing carryovers/ contamination s from previous injections or samples and thus the quality of analysis is not compromised. More importantly, the Waters Alliance Separations Module has a low total system volume, <650 mL, with excellent performance. This system produces accurate gradients with good eluent mixing, and excellent retention time reproducibility. Unit to unit consistency has been designed into the Alliance, making gradient methods transfer easier These HPLC Systems are used to transfer an HPLC method for impurities analysis from an Alliance System, successfully replicating the quality of chromatographic separation. The relative retention times are comparable, eliminating the need to make manual dwell volume adjustments for effective method transfer

Transfer of USP Assay for Quetiapine Fumarate Across Different LC Systems

In this study, the USP assay method for quetiapine fumarate, an anti-psychotic drug, will be evaluated on a range of systems including an Alliance HPLC System, an ACQUITY Arc UHPLC System and an ACQUITY UPLC H-Class PLUS System. The USP method for quetiapine fumarate was successfully transferred across three systems: an Alliance HPLC System, an ACQUITY Arc UHPLC System, and also an ACQUITY UPLC H-Class PLUS System. All three instruments were able to reproduce the percent of quetiapine fumarate in an unknown sample and meet system suitability requirements, including resolution, tailing factor, and repeatability specifications required by the USP quetiapine fumarate assay method.

Scaling of a USP Assay for Quetiapine Fumarate Across Different LC Systems

In this study, the Waters Columns Calculator will be used to scale the USP monograph assay method for quetiapine fumarate an anti-psychotic drug, to smaller particle sized columns.

Utilizing the method scaling guidelines within the USP General Chapter <621>, traditional isocratic HPLC methods are capable of being scaled to columns with smaller particle sizes and shorter lengths. Scaling a USP method enables the use of modern chemistries and LC hardware to deliver improved throughput with decreased solvent consumption all while providing accurate and reproducible chromatographic data. The scaled modifications for the USP quetiapine fumarate assay method enabled the original method run time to be reduced by 57% for a 3 μm column on an ACQUITY Arc UHPLC System and 87% reduction for the 1.7 μm column on an ACQUITY UPLC H-Class PLUS System. The scaled methods maintained the same or improved chromatographic performance in terms of resolution, peak tailing, and retention time and peak area RSD.

This poster presents the results of experiments that were carried out to qualify POU polishers.
