We developed an easy transformation method called "easyJanus" to facilitate the gene deletions, additions, or point mutations in S. pneumoniae. The use of synthetic DNA saves time, reduces the need for multiple PCR reactions, and increases the precision of gene editing.

Plasmid construction
I've been struggling to make a particular plasmid for months, using DNA produced by competing services. This has been difficult because of extensive repetitive elements, which Twist's competitors can't synthesis in one fragment. Assembling these repetitive elements has been very difficult, and I have not been successful with this. Twist was able to synthesize the DNA I need all in one piece. They have done this faster and cheaper than the other companies I have used in the past. Every colony I sequenced has the correct DNA insert - I haven't yet seen any errors in the DNA I received from Twist. This has let me finally make the plasmid I was previously struggling with, but faster, easier, and cheaper than I had imagined could be possible. My only regret is that I didn't find out about Twist earlier!
